Single-cell Proteomics by Mass Cytometry

We provide state-of-the art Mass Cytometry (CyTOF™, Fluidigm Inc.) analysis to the entire Swedish research community. The facility belongs to the Affinity proteomics platform within SciLifeLab and consists of two nodes, one in Stockholm and one in Linköping. Together we perform standardized Mass Cytometry experiments on customer provided samples using validated panels of metal-coupled antibodies. We also perform technology development of new experimental assays and reagents as well as novel data analysis tools.


Mass Cytometry

Mass Cytometry allows for many more antibodies to be combined and resolved in individual cells as compared to traditional fluorescence-based cytometry. This is thanks to the use of antibodies coupled to isotopically purified mass tags, each with a unique mass, that can be quantified in individual cells using an Inductively-Coupled Plasma Mass Spectrometer (ICP-MS), adapted to the analysis of individual cells in suspension (CyTOF2™, Fluidigm, Inc.). In this way, about 40-simultaneous antigens can be quantified in individual cells at a rate of about 500-1000 cells/second.

Validated Antibody Panels

Mass cytometry provides a powerful way to better understand heterogeneous cell suspensions, like whole-blood or peripheral blood mononuclear cells (PBMC) samples. Both mouse and human cells can be analyzed using one of our validated antibody panels.

Sample Preparation for Mass Cytometry (CyTOF)

Human & Mouse samples

Human Peripheral blood mononuclear cells (PBMCs) or mouse cell suspension should be frozen using a cryoprotective media containing 10% dimethyl sulfoxide (DMSO) and 90% fetal bovine serum (FBS) and stored in -80oC or Liquid Nitrogen. The samples should be transported (locally)/shipped (outside Stockholm) in a box with Dry ice or Liquid Nitrogen containers to our facility.
